How deficiencies in 2 genes synergize to halt formation of gut nervous system

KANSAS CITY, MOMutations in single genes can cause catastrophic diseases, such as Huntington's Disease or sickle cell anemia. However, many conditions, including cancer, diabetes and birth defects are multigenic, arising from the collective failure of the function of more than one gene.

Researchers know that mutations in at least twelve individual genes are associated with the congenital defect Hirschprung Disease (HSCR), in which children are born lacking nerves that innervate the large intestine. Now two companion studies published in Human Molecular Genetics by Paul Trainor, Ph.D., Investigator at the Stowers Institute for Medical Research, identify a new gene associated with HSCR and show how the migration of cells that form the gut nervous system is impeded when the combined doses of two candidate genes are low. Understanding the genetic basis of HSCR offers hope for better diagnostics and treatment for it and other developmental defects caused by failure of cell migration.

The cells that go awry in HSCR are a subset of what are called neural crest cells, embryonic cells that spring from the developing brain and spinal cord in mice or humans and then travel long distances to form, among others, structures in the face and heart, smooth muscle, and neurons of the peripheral nervous system, including those that innervate the gut.

Trainor has been interested in neural crest cells since he was a graduate student, often focusing on developmental defects caused by their malfunction. "Neural crest cells have to be born in the right place, migrate an incredibly long distance, survive the migration, multiply and then differentiate into a mature cell type," says Trainor, who until recently was primarily interested in neural crest-related craniofacial anomalies. "In Hirschprung Disease they don't make it to the end of the large intestine."

As a result, the affected portion of the intestine cannot undergo peristaltic movements and waste products accumulate in the gut. Although surgery to remove the affected part of the gut improves the condition, some children are left with lasting complications including constipation, dehydration and poor nutrient absorption.

Both Human Molecular Genetics papers employ imaging techniques to visualize and track neural crest migration in mice mutant in HSCR candidate genes. The earlier study, published in January of 2012, reported that mice missing one of their two copies of a gene called Tcof1 not only produced fewer neural crest cells but that colonization of the gut by the remaining cells proceeded slowly compared to normal mice. Despite that delay, lagging cells eventually reached the end of the intestine before mice were born, just in time to innervate the gut properly.

"That paper showed that neural crest cells don't seem to have a defined time window to colonize the entire gut as long as they do it by birth," says Trainor, suggesting that for humans this is a good thing in terms of potential intervention. "If an embryo looked like it was going to have Hirschprung's characteristics, that did not necessarily mean it would end up with that condition."

If the first study showed that loss of a little Tcof1 had unusual but non-debilitating effects on gut innervation, the second paper, published in January of 2013, revealed what happens when you genetically chip away at the process. In the latter, the researchers eliminated one copy of Tcof1 along with one copy of another gene known to govern neural crest formation and proliferation, Pax3. Double mutant mice showed serious defects: fewer neural crest cells migrated out of the embryonic spinal cord, and the ones that remained underwent increased cell death as they traveled toward the gut. As a result, near the end of embryogenesis, Pax3/Tcof1 double mutant mice lacked the normal complement of neurons in the large intestine, a condition mimicking human HSCR.

"This is a much more dramatic result than the effect you might see following decreases in either gene alone," says Amanda Barlow, Ph.D., a former Senior Research Associate in the Trainor lab and first author of both studies. "It's like having two hammers which deliver two blows."

Some molecular biologists might alter that metaphor slightly, to perhaps a hammer and crow bar, as Tcof1 and Pax3 are very different proteins. Pax3 is a "logical suspect", a transcription factor that binds DNA and turns target genes known to stimulate cell proliferation off and on. By contrast, Tcof1 resides in a specialized cellular compartment called the nucleolus where the machinery that synthesizes all cellular proteins is constructed. How loss of a little of these strange bedfellows synergizes to promote developmental defects remains unknown. Trainor says it's likely a threshold effect: losing a little Tcof1 might debilitate cellular growth just enough to make neural crest cells vulnerable to death if Pax3 doses were also compromised.

Barlow, now an Associate Scientist at University of Wisconsin, agrees, noting that researchers are increasingly aware that genetic interactions among seemingly unrelated genes are likely the norm, not the exception. "No gene exists in isolation," she says. "To make an embryo the activity of many different pathways have to be integrated. There's going to be a lot of cross-talk."

###

In addition to Trainor and Barlow, Mike Dixon, Ph.D., and Jill Dixon, Ph.D., of the University of Manchester in the UK contributed to both papers. Long-standing collaborators with Trainor, the Dixons were part of the team that first identified TCOF1 as responsible for Treacher Collins syndrome craniofacial anomalies, and they made the Tcof1 mouse model used in the these investigations.

The study was funded by the Stowers Institute for Medical Research, the March of Dimes, National Institute of Dental and Craniofacial Research (RO1 DE 016082-01) the National Institutes of Health (P50 DE 016215), the Healing Foundation and the Medical Research Council of the UK.

Opinions on Red Hook? - USVI Moving Center

First off, hello everyone. This is my first post here, though I have been enjoying all of your insight and searching on this fomum for about 60 days now. It's so nice to see so many people willing to share their time in helping others who want to be, or are in the process of becoming, their neighbors. Have actually enjoyed reading AandAtoVI's saga - like they were, we were initially focused on St Croix but after a PMV there are targeting St Thomas.

So after visiting St Croix a few weeks ago and seeing several houses (and having close to 20 hours on the St Croix MLS, using a STX Realtor), we decided STX is just not for us. It was just way to quiet and seemingly desolate for us. We are 'town' people - we like to go out to restaurants all the time, we like activity, and while I always considered us laid back, compared to Crucians we are apparently quite uptight ;-)

It was a very good experience, however, and we've been searcing the St. Thomas MLS since we got back 2 weeks ago and really decided that we really liked the Red Hook area, and Nazareth. 90+% of the homes that meet our selection criteria are in Mahogony Run, Frenchman's Bay, or Nazareth -- but while I LOVE golf (live on a course in MI) it seemed that was really all that area had going on, it's cooler, and had mildew issues. Frenchman's Bay looked nice, but not "neighborhoody" enough for us. We have a 4 year old and being in an area where he can make friends is very important to us (we want to make friends too of course). This is one of the biggest reasons that the Red Hook area was so appealing to us - we'd like to have a 'town' (I know it's not technically a city) quite close by to hang out in and enjoy life in that is safe to be in after the sun sets, and almost anywhere you are in Nazareth you are within walking distance to a fabulous beach.

Any opinions about the area? We're going for our STT PMV in less than 2 weeks, and are going to stay at Secret Harbor. My wife is in the process of selecting a STT Buyer's Agent, but if anyone has a strong recommendation please pipe up - we'd want someone familiar with the Red Hook area, and with single homes in the median range for the areas (not condos). We have 3 houses we're planning to see when we go (unless you all persuade us otherwise about the area) - would love any comments - again a big part for us is having kids nearby for our four year old to play with, so knowing what parts of the area are more family style versus empty nesters would be a big help.

BTW, I know most new folks are told not to buy something, and to rent first for a year, but in our case we've seen every house the the MLS on the island 5 times over, and there are really only 2 or 3 we're interested in. Given that we're only going to have a new house that fits us come on line every 4 months or so, we don't mind taking a risk and buying before we move. We'd plan to spend our summers in Michigan anyway (Michigan is absolutely wonderful in the summer, and the sun doesn't set until 9:30 in June and July), so if it didn't pan out we would just rent the house all full-time instead of just in the summer. In terms of job we're all set; as long as we like STT I'll be starting a new company based in the east end of St. Thomas in the fall.

Thanks first off for all of your past comments - I've done a lot of post searches here and really enjoyed your perspecives and candor (I don't mind negative comments at all). Appeciate very much any opinions you all have. We've lived our whole lives in a 50 mile circle and instead of "when we retire we'll......" we're trying to take a leap and do it now when were 40 and still have [hopefully] most of our lives ahead of us.

Cheers,

Chris (and Nicole)

PKK guerrillas made no official ceasefire declaration: spokesman

BAGHDAD (Reuters) - Kurdish militants have made no official declaration of any ceasefire for moment, a PKK guerrilla spokesman said in response to a media report the group will halt hostilities in Turkey as part of a fledging peace process.

The mainstream Hurriyet newspaper reported on Tuesday that 100 fighters from the Kurdistan Workers Party or PKK will hand in their weapons and leave Turkey as part of initial attempts to end their 28 years of insurgency.

"The PKK officially has made no such declaration for the moment," spokesman Roj Welat said by telephone. "There is no such information in our hands."

Seeking Roleplaying Partners Long-Term

I am new to this forum, and looking for some new role plays and partners.

About Me:
I have no problem playing either gender. There are some plots where I prefer a certain gender, but for the most part, it doesn't matter. I don't mind doubling up - if the plot calls for it. I also like tossing in NPC and background characters, for spice and what-not. I'm a high-casual to advanced writer. I like 3+ paragraphs per post typically. And a paragraph is usually between 4-6 sentences, not two, and certainly not your entire post. Just sayin'. Usually, I return what I'm given. Also, please write in third person, past tense. It makes it less painful to read. If you don't, I won't reply. As an example, that's "he went into the market" not "he goes into the market" and certainly not "I am going into the market."

As far as maturity, I have no problem with swearing, gore, violence, drinking and drug usage, that sort of stuff.

As a warning, I'm a very fickle being. Within one to five posts, I can tell if a RP will last. If the spark isn't there, it just isn't. People have likes and dislikes, and I have no problem bidding farewell if it isn't working for either of us. You should, too.

Now, for how often to post. I don't really have a time table. Sometimes, I post often. Sometimes, I'm really just not feeling it. It just depends on my mood, and how excited I am over the rp. I likely won't post every day. Feel free to nag me. Bitch, moan, throw a hissy. It works. But, be forewarned I just won't be doing it every day.

Ideas and What I Like:

Vampire Hunters:
Right now, I am REALLY, REALLY craving a vampire/werewolf hunter RP. Not the typical hunterxvampire, but one in which our two characters are hunters. I would prefer it take place in the early/mid 1800s, though his is completely negotiable. I want it to focus on the relationship between the two hunters, evolving over time. A good buddy story, that happens to include vampires and werewolves and the occasional (or not so occasional) explosion. My character for this would be female, if you would like to know about her feel free to ask. The RP could have light-steampunk for weaponry and such, but nothing over the top.

Bonnie and Clyde/Natural Born Killers:
The classic tale of two lovers on the lam, running from the law and upping the bodycount as they go. I do want a genuine soulmate connection between the two characters, not a transference of lust and adrenaline misplaced as love. Also would like a sense of nature and nurture, one of the characters coming from the typical broken, possibly abusive home, and the other from a nice white-picket-fence upbringing.

In a world where people can't make the simplest relationship work, they have a "do or die" romance of Shakespearean magnitude. They have created a world where only two exist, and anyone who inadvertently enters that world is killed. Two deranged beings that somehow found their counterpart, and have a bloody way of showing their affections to the world. The plot is malleable depending on the characters, and plenty open for discussion. But, I also have a basic plot that can be used.

I like:
Vampires
Werewolves
Urban Fantasy
Medieval Fantasy
Dystopian Futuristic
Supernatural-hunters
Zombie-Apocalypses
Post-Apocalyptic
Modern/realistic (dependent on plot)
Historical/Pirate
Greek/Roman/Egyptian/Atlantean/Barbaric
Mutants
Elementals
Horror (would especially be interested in a 'cursed carnival' if a good plot could be devised.)

Fandoms:
Repo! The Genetic Opera
Devil's Carnival
Legend of the Seeker

Will edit as more comes to mind.

Xerox Travel Scanner 150

The Xerox Travel Scanner 150 ($159.99 direct) is a featherweight, USB-powered portable scanner geared toward travelers who need to scan documents, receipts, and/or business cards while on the road. It can scan to a number of destinations, including cloud-based ones, in a variety of document formats, but it lacks an automatic document feeder (ADF) and is limited to simplex scanning.

The Travel Scanner 150 is very compact and lightweight, measuring 1.5 by 12.4 by 2 inches (HWD) and weighed in at just 11.2 ounces. As it is powered from a computer over a USB connection, there's no power adapter to lug. Unlike many portable scanners, including two Editors' Choice models?the Xerox Mobile Scanner (4 stars) and the Visioneer Mobility (4 stars) ?it can't operate independently of a PC. The Travel Scanner 150 includes a good selection of bundled software: the Visioneer OneTouch scan utility; Visioneer Acuity for scan optimization and quality enhacement; Nuance PaperPort Professional 12 for document management; Nuance OmniPage Pro for OCR; Nuance PDF Converter; and NewSoft Presto! BizCard 5 for business cards.

Scanning
The OneTouch utility's default pre-programmed scan profile is Document (PDF), which scans to 200-ppi black-and-white image PDF and saves it to a OneTouch folder in PaperPort. The OneTouch interface, accessible from an icon on the Start bar (we tested it on a Windows Vista system) offers a variety of additional scan profiles, which can be enabled or modified through the interface. They include but are not limited to Searchable PDF, OCR, legal-sized or A4 documents, receipts, business cards, and bank checks. It can scan to image PDF, Searchable PDF, BMP, TIFF, and JPEG, as well as to DOC, RTF, and other document formats. You can change resolution (it goes up to 600 ppi), mode (black-and-white, grayscale, or color), and more.

It will also scan to Cloud-based services? Google Docs, Evernote, FilesAnywhere, OfficeDrop, and DropBox ?by means of OneTouch links, which are downloadable for free from Visioneer Connect. They can be preset with login credentials entered and destination folder selected.

You can also scan from PaperPort, or from most any program that offers a scan command, thanks to the enclosed Twain and WIA drivers as well as Visioneer's own DriverPlus driver.

Scanning is simple and straightforward?just insert the document in the slot, and the scanner should grab it and feed the paper through. It says on the Xerox data sheet for the Travel Scanner 150, "? if you can insert a dollar bill into a change machine, you have the scanning operation mastered ?" That's true enough, but just as a change or vending machine won't always accept your dollar bill on the first try, ?at times the Travel Scanner 150 proved finicky and had to be coaxed into accepting a sheet.

Results
The Travel Scanner 150 did well on our OCR testing, reading our Arial font test page perfectly down to 5 points except for two minor errors (one spurious italicization and one spurious capitalization), and our Times New Roman font test page perfectly down to 8 points, with a couple of minor errors at 6 points. It did well in business-card scanning with BizCard, with about half of the cards not showing any errors, and most of the others just one or two.

The Xerox Travel Scanner 150 is a portable document scanner for the business traveler that needs to scan receipts and other documents while on the road. It's featherweight and compact, and can scan to a variety of destinations and document formats, including searchable PDF. Its features are basic: It supports simplex (one-sided) scanning only, lacks an automatic document feeder (ADF), and it requires a USB connection to a computer to operate.

If you need a portable scanner that adds an ADF and supports duplex scanning, consider springing for the Editors' Choice Canon imageFormula P-150 Scan-tini. The battery-powered Visioneer Mobility and Xerox Mobile Scanner, also Editors' Choices, each support scanning to mobile devices?the former over a cable and the latter via an Eye-Fi Wi-Fi card. But for many travelers, simplex scanning of receipts, business cards, and other short documents to a laptop via USB cable will suffice, and the Xerox Travel Scanner 150 provides an easy, effective, and cost-effective solution.

Cities affect temperatures for thousands of miles

Jan. 27, 2013 ? Even if you live more than 1,000 miles from the nearest large city, it could be affecting your weather.

In a new study that shows the extent to which human activities are influencing the atmosphere, scientists have concluded that the heat generated by everyday activities in metropolitan areas alters the character of the jet stream and other major atmospheric systems. This affects temperatures across thousands of miles, significantly warming some areas and cooling others, according to the study this week in Nature Climate Change.

The extra "waste heat" generated from buildings, cars, and other sources in major Northern Hemisphere urban areas causes winter warming across large areas of northern North America and northern Asia. Temperatures in some remote areas increase by as much as 1 degree Celsius (1.8 degrees Fahrenheit), according to the research by scientists at the Scripps Institution of Oceanography; University of California, San Diego; Florida State University; and the National Center for Atmospheric Research.

At the same time, the changes to atmospheric circulation caused by the waste heat cool areas of Europe by as much as 1 degree C (1.8 degrees F), with much of the temperature decrease occurring in the fall.

The net effect on global mean temperatures is nearly negligible -- an average increase worldwide of just 0.01 degrees C (about 0.02 degrees F). This is because the total human-produced waste heat is only about 0.3 percent of the heat transported across higher latitudes by atmospheric and oceanic circulations.

However, the noticeable impact on regional temperatures may explain why some regions are experiencing more winter warming than projected by climate computer models, the researchers conclude. They suggest that models be adjusted to take the influence of waste heat into account.

"The burning of fossil fuel not only emits greenhouse gases but also directly affects temperatures because of heat that escapes from sources like buildings and cars," says NCAR scientist Aixue Hu, a co-author of the study. "Although much of this waste heat is concentrated in large cities, it can change atmospheric patterns in a way that raises or lowers temperatures across considerable distances."

Distinct from urban heat island effect

The researchers stressed that the effect of waste heat is distinct from the so-called urban heat island effect. Such islands are mainly a function of the heat collected and re-radiated by pavement, buildings, and other urban features, whereas the new study examines the heat produced directly through transportation, heating and cooling units, and other activities.

The study, "Energy consumption and the unexplained winter warming over northern Asia and North America," appeared online January 27. It was funded by the National Science Foundation, NCAR's sponsor, as well as the Department of Energy and the National Oceanic and Atmospheric Administration.

Hu, along with lead author Guang Zhang of Scripps and Ming Cai of Florida State University, analyzed the energy consumption -- from heating buildings to powering vehicles -- that generates waste heat release. The world's total energy consumption in 2006 was equivalent to a constant-use rate of 16 terawatts (1 terawatt, or TW, equals 1 trillion watts). Of that, an average rate of 6.7 TW was consumed in 86 metropolitan areas in the Northern Hemisphere.

Using a computer model of the atmosphere, the authors found that the influence of this waste heat can widen the jet stream.

"What we found is that energy use from multiple urban areas collectively can warm the atmosphere remotely, thousands of miles away from the energy consumption regions," Zhang says. "This is accomplished through atmospheric circulation change."

The release of waste heat is different from energy that is naturally distributed in the atmosphere, the researchers noted. The largest source of heat, solar energy, warms Earth's surface and atmospheric circulations redistribute that energy from one region to another. Human energy consumption distributes energy that had lain dormant and sequestered for millions of years, mostly in the form of oil or coal.

Though the amount of human-generated energy is a small portion of that transported by nature, it is highly concentrated in urban areas. In the Northern Hemisphere, many of those urban areas lie directly under major atmospheric troughs and jet streams.

"The world's most populated and energy-intensive metropolitan areas are along the east and west coasts of the North American and Eurasian continents, underneath the most prominent atmospheric circulation troughs and ridges," Cai says. "The release of this concentrated waste energy causes the noticeable interruption to the normal atmospheric circulation systems above, leading to remote surface temperature changes far away from the regions where waste heat is generated."

UFC on Fox 6 results: Demetrious Johnson retains title, ?Rampage? loses in Octagon finale

CHICAGO -- The UFC's stop in the Windy City had two great knockouts, a champion holding onto his belt, and the last fight of a one-time champ.

Despite a strong start by John Dodson, Demetrious Johnson held onto his championship belt with a unanimous decision. The judges saw it 48-47, 49-46, 48-47 for Johnson.

Unsurprisingly, Johnson and Dodson fought a fast-paced first two rounds. Johnson tried to slow things down in the first round with a takedown. While he did get Dodson to to the ground, the challenger popped back to his feet quickly.

Dodson dropped Johnson twice in the second round, and shook off Johnson's take down attempts with a great sprawl. Johnson seemed to have a hard time even getting close to Dodson to land a punch.

[Related: T.J. Grant and Ryan Bader shine at UFC on Fox 6]

A Johnson knee in the fourth round caused a small fight stoppage. Dodson's hand was on the ground as Johnson threw a knee to Dodson's head, which is an illegal strike. The bout was stopped as doctors checked Dodson's eye and Johnson was warned. The fight went on, but not without plenty of boos from the crowd in Chicago.

When the fight restarted, it was all Johnson. He controlled Dodson against the cage, and threw knees that busted up Dodson's face. Johnson was able to get a takedown at the beginning of the fifth round, too. Though boos rained down, Johnson kept the fight against the cage.

Later, the crowd got behind him as Johnson elevated, tightened his legs around Dodson's torso and threw elbows. It was a creative move that likely could only happen in the flyweight division. Johnson finished the round with Dodson against the cage, and knee after knee after knee to the body and face.

Quinton 'Rampage' Jackson vs. Glover Teixeira

If this really was Quinton "Rampage" Jackson's last fight in the UFC, it wasn't one to remember. He was outstruck and outwrestled by Glover Teixeira throughout their bout. Teixeira took the fight 30-27, 30-27, 29-28 on the judges' cards.

Teixeira's first round was his best, as he was able to take Jackson down early and mount him and take his back. Late in the round, Teixeira knocked Jackson down with a punch and looked close to finishing, but wasn't able to end the fight.

The rest of the fight featured a worn out Jackson trying to avoid Teixeira's takedown attempts unsuccessfully. Jackson threw some big punches, and even jawed at Teixeira during the bout, but he wasn't able to score any big offense.

While this fight will be remembered for being Jackson's last UFC bout, Teixeira scored his third straight UFC win on Saturday night. He looked impressive against the former champ.

Anthony Pettis vs. Donald Cerrone

Anthony Pettis won his much anticipated bout with Donald Cerrone by landing big, memorable strikes. Pettis was getting the better of their striking exchanges early, which set him up for a big finish. Pettis threw a kick that landed hard on Cerrone's body, then finished with a punch. Cerrone fell to the ground in a heap, and the fight was stopped at 2:35 in the first round.

Pettis lost his first fight in the UFC after coming from the WEC as the champ, but he's been winning ever since. He has wins over Joe Lauzon, Jeremy Stephens and now Cerrone. After the fight, he made a pitch to UFC president Dana White.

"I want my title shot. I should have had it years ago. People say I can?t wrestle because of the Guida fight, but I had a serious shoulder injury. Now I am 100 percent. No one can do in the Octagon what I can do."

While lightweight champion Benson Henderson has a fight set up with Gilbert Melendez, a shot for Pettis isn't unwarranted. It would be a rematch of their WEC title fight, which Pettis won in the promotion's final fight.

Ricardo Lamas vs. Erik Koch

Lamas won the first round by constantly pressuring Koch against the cage. It wasn't exactly the most thrilling round to have start a card on network television, but it led to a thrilling second round. Lamas took advantage of Koch's slip, and then finished the fight with nasty, nasty ground and pound. Lamas started with elbows, then started with strikes that cut open Koch's face. Koch had no answer for Lamas' relentless strikes, and the bout was stopped at 2:32 in the second round.

Lamas thought his performance warranted a title shot.

"I?ve beat Cub Swanson, I?ve beat Hioki and now I?ve beat Koch. All those guys were supposed to be fighting for the title at one point. I beat them all. We?re all here to be world champ and I am no different. I want the winner of Aldo vs Edgar next week. I don?t care who wins, I want the winner."

Venezuela prison riot kills 61, government mum

URIBANA, Venezuela (Reuters) - A prison riot in southwestern Venezuela killed 61 people, a hospital official said on Sunday, although the government has refused to give an official death toll in the bloody standoff that highlighted chaos in the country's jails.

The violence took place on Friday at the Uribana jail near the city of Barquisimeto. Relatives who gathered outside the jail fumed at the lack of information from authorities who have started transferring prisoners to other facilities but have not confirmed how many were killed.

The rioting took place at a time when cancer-stricken President Hugo Chavez remains in Cuba receiving treatment and with Vice President Nicolas Maduro - the notional head of state - visiting the socialist leader in Havana and then traveling to a summit in Chile.

The riot was the fourth high-profile conflict in 18 months in a prison system that houses three times the number of inmates it was designed to hold. Critics say it is controlled by gangs with ready access to machine guns and even hand grenades.

"We're suffering here, and the government is saying nothing," said Josefina Ramirez, 36, whose 25-year-old husband was inside. "We want Chavez to come here to give us news. We want an answer."

Chavez has not been seen or heard from in 45 days, which has spurred criticism from opposition leaders that the country is effectively without a president. Allies insist he is fully carrying out duties.

Ruy Medina, director of the Barquisimeto Central Hospital, put the death toll at 61.

A hand-written list posted on a barbed wire fence outside the Uribana jail listed 20 dead and 104 injured. Outside the nearby morgue, where hearses lined up on Saturday to collect bodies, a similar list showed 24 dead.

It was not known who posted the lists. A prisons ministry official did not respond to requests for comment. Amid the silence, rumors circulated among family members at the prison gates that the death toll had reached as high as 400.

'BUNCH OF DEAD PEOPLE'

"There's a bunch of dead people tossed on the ground in there, the government doesn't want to take them out to avoid showing the reality," said Veronica Chavez, whose husband told her he was being transferred to another prison but did not know which. She called the list outside the prison "a lie."

Maduro vowed a full investigation of the incident in pre-dawn comments on Saturday just after arriving back in Venezuela from Havana. Within hours, he left to meet with Latin American and European dignitaries at a summit in Chile.

Brazilian President Dilma Rousseff scrapped her agenda at that summit and returned home after a nightclub fire killed at least 232 people.

Prisons Minister Iris Varela in a news conference said the riot was triggered by reports in the opposition media about plans to search the prison to remove weapons. "It seems as though their thirst for blood and terrible things will never be quenched," she said, referring to media.

Varela said reports had exaggerated the number of dead by including prisoners whose bodies showed signs they died several days before the incident - comments that critics seized as further evidence of the cruelty of the penitentiary system.

"They're not dogs, they're not animals, they're people like us," said Angelia Ibarra, 42, seeking information about her 25-year-old son inside. "The true animals are the government people."

Venezuelan jails have been increasingly in the headlines because of repeated shootouts and riots as well as conditions that are both outrageously cruel and downright surreal.

Inmates refusing to be transferred out of a Caracas prison battled security forces in May for days as smoke rose above the compound and shots rang out. Chavez later said he called from Cuba, where he was receiving medical treatment, and spoke with one of the inmates to help negotiate an end to the standoff.

Local media last year published a story about a nightclub called Disco Tokio that held a Mother's Day party that featured musical groups flown in from Colombia and Puerto Rico. The club was located inside the Tocoron jail.

An online animated series about jail violence called "Jail or Hell," produced by a former inmate, has drawn a following among Venezuelans captivated by the chaos of the prisons.

Unpopular Dog Names: Monikers For Pets That Are No Longer Cool

From Vetstreet's Kristen Seymour:

Just as today?s second-grade classrooms are no longer filled to the brim with Jennifers and Jasons, the dog names that were most popular a decade or two ago are no longer at the head of the class.

After checking out the most popular and trendiest puppy names, we decided to try something different and determine which names are on the other end of current trends. Using Vetstreet?s data on close to 1.26 million dogs, we took a peek at which names have fallen in popularity since 2003. Here are the 10 male and 10 female dog names that have fallen the farthest in the past decade.

List and captions courtesy of Vetstreet

  • #10 Sammy (Male) / Shadow (Female)

Sammy was the No. 24 most popular male dog name a decade ago and fell to No. 66 last year. It?s worth noting that Sam is also losing popularity, falling from No. 14 to No. 35 over the same period of time. Shadow, which came in as the No. 51 most popular female dog name in 2003, has now fallen to No. 87.

  • #9 Smokey (Male) / Madison (Female)

Smokey hasn?t fallen out of the top 100, but after going from No. 43 to No. 87 over the last decade, it still earned a spot on this list. Madison, the No. 62 female dog name in 2003, dropped out of the top 100 in 2009. Its nickname, Maddie, has also lost popularity but at a slower pace.

  • #8 Luke (Male) / Sheba (Female)

Although Luke was the No. 56 most popular name for male dogs in 2003, it fell out of the top 100 in 2012. Sheba, a name that fell off the list of most popular female dog names in 2009, came in at No. 61 back in 2003.

  • #7 Scooby (Male) / Casey (Female)

Scooby (Dooby Doo), where are you? Not in the top 100 anymore. After coming in at No. 53 in 2003, the name of one of pop culture?s favorite pups fell out of the top 100 in 2008. Casey, which was the No. 58 most popular female name a decade ago, also fell off the most popular names list in 2008.

  • #6 Rusty (Male) / Heidi (Female)

Rusty had a home on the list of 20 most popular names a decade ago, coming in at No. 16, but last year it dropped to No. 63. Heidi is still in the top 100 most popular female dog names ? but only just, coming in at No. 92 after holding the No. 50 spot in 2003.

  • #5 Joey (Male) / Lucky (Female)

Joey, the No. 52 most popular male dog name in 2003, fell out of the top 100 in 2012, earning it the No. 5 spot on this list. Lucky was the No. 56 most popular female dog name in 2003 but dropped off the list in 2010.

  • #4 Spike (Male) / Katie (Female)

Spike has fallen drastically in popularity in the past decade, going from No. 29 in 2003 to No. 81 in 2012. Katie saw a similar drop, beginning at No. 33 a decade ago, then falling to No. 82 last year.

  • #3 Rudy (Male) / Misty (Female)

Rudy was in the top half of most popular names in 2003, coming in at No. 47, but fell out of the top 100 last year. After holding the No. 48 spot in 2003, Misty dropped off the 100 most popular names list in 2011.

  • #2 Dakota (Male) / Holly (Female)

Dakota, the No. 43 most popular male dog name in 2003, comes in second on our list after dropping out of the top 100 in 2009. Holly, the second least trendy female dog name, has a similar story, claiming No. 45 in 2003 and falling off the list of most popular names in 2012.

  • #1 Scooter (Male) / Brandy (Female)

Scooter earned the top spot on this list by declining the most in popularity over the past decade, falling from No. 34 on the list of most popular male dog names in 2003 to No. 98 in 2012. Brandy takes No. 1 for females on the least popular list, starting at No. 42 in 2003 and falling out of the top 100 in 2011.

Trial under way in LA hip joint replacement suit

LOS ANGELES (AP) ? A California jury heard opening statements Friday in a lawsuit that accuses Johnson & Johnson of knowingly marketing a faulty hip implant that lawyers say left thousands of people with crippling problems or in need of replacement surgeries.

The fraud and negligence suit is the first of thousands of similar cases to reach trial in the United States that involve an all-metal ball-and-socket hip joint that was pulled from the market two years ago.

A lawyer for plaintiff Loren Kransky showed jurors pictures from a surgery in which black material could be seen in the patient's hip socket. Attorney Michael Kelly said the material was from pieces of metal that had flaked off of the implant.

Kelly also played a segment of a doctor's audio deposition in which he said he feared that if the material wasn't removed, Kransky would have died. The pieces of metal were causing a form of metal poisoning, he said.

Kransky, a former North Dakota prison guard, sought hip replacement to relieve arthritic pain. He received the implant in 2007 and has since has had it replaced. He listened to opening statements Friday from a front row seat.

A lawyer for Johnson & Johnson's subsidiary, DePuy Orthopedics Inc., which manufactured the metal implant, said Friday that the 64-year-old Kransky had many pre-existing medical ailments. Attorney Alexander Calfo presented a list of 16 major diseases, including kidney cancer, diabetes, high blood pressure and vascular disease as well as exposure to Agent Orange during the Vietnam War.

"Mr. Kransky did not get worse because of the (implant) and did not get better when it was removed," he said.

He added: "Hip surgery is not perfect. No material to this very day has proven to be perfect."

Calfo said the metal implant had been designed to improve on plastic and ceramic implants.

The artificial hip socket was sold for eight years to some 35,000 people in the U.S. and more than 90,000 people worldwide. New Brunswick, N.J.-based Johnson & Johnson stopped making the product in 2009 and recalled it the next year.

However, documents unsealed in the court case last week indicated that Johnson & Johnson officials were aware of problems with the device at least as far back as 2008.

Also, according to a deposition from a DePuy official, a 2011 company review of a patient registry concluded that more than one-third of the implants were expected to fail within five years of their implantation.

Johnson & Johnson has put aside around $1 billion to deal with the costs of the recall and lawsuits.

Last year, British experts at the world's biggest artificial joint registry said doctors should stop using metal-on-metal hip replacements after a study found that, after five years, about 6 percent of people who had used them needed surgery to fix or replace them.

That compares with just 1.7 to 2.3 percent of people who had ceramic or plastic joints.

Kelly said he will ask the jury to assess punitive damages at the end of the trial "to send a message to the defendants who failed to share with doctors what they knew."

He said that when DePuy Orthopedics learned the device they manufactured might be defective, "They acted in a manner that was indifferent."

Jindal: 'Stop Being The Stupid Party'

By MICHAEL FALCONE ( @michaelpfalcone )

NOTABLES:

BREAKING - OBAMA TO ANNOUNCE CHIEF OF STAFF: From the White House: "This afternoon, the President will introduce Denis McDonough as the new White House Chief of Staff. The President will make this personnel announcement at the White House at 12:10 PM in the East Room. Denis has been one of the President's closest and most trusted advisors for nearly a decade, dating back to when he helped set up then Senator's Barack Obama's Senate office. He has relied on his intellect and good judgment ever since- most recently as a member of the President's national security team. In that capacity, Denis has played a key role in all of the major national security decisions - from ending the war in Iraq to winding down the war in Afghanistan; from our response to natural disasters in Haiti and Japan; to the repeal of 'Don't Ask, Don't Tell.'"

FORMER DEFENSE SECRETARY, NATIONAL SECURITY OFFICIALS ENDORSE HAGEL. Chuck Hagel, President Obama's controversial secretary of Defense nominee, received a boost on Thursday, ABC's Jonathan Karl reports: Former Secretary of Defense Robert Gates - who served under both George W. Bush and Barack Obama - has signed on to a letter with a bi-partisan group of 12 other former top national security officials expressing his "strong endorsement" of Chuck Hagel for Secretary of Defense. The list also includes several top national security officials for Republican presidents: Brent Scrowcroft (Bush 41), Mel Laird (Nixon), George Schultz (Reagan), Colin Powell (Reagan, Bush 41 and Bush 43), Robert McFarlane (Reagan). It's a pretty formidable list. http://abcn.ws/WS4iWP

HAPPENING TODAY: Vice President Joe Biden, Secretary of Homeland Security Janet Napolitano, Secretary of Health and Human Services Kathleen Sebelius, Deputy U.S. Attorney General Jim Cole, and other Obama administration officials will hold a roundtable discussion on reducing gun violence at Virginia Commonwealth University in Richmond, Va. Sen. Tim Kaine, D-Va., and Rep. Bobby Scott, D-Va., will also attend. According to the White House, the session will include experts who have worked on gun safety issues in the wake of the Virginia Tech shooting.

THIS WEEK ON 'THIS WEEK': After Hillary Clinton faced off with Congress on Benghazi, Sen. John McCain, R-Ariz., and Sen. Robert Menendez, D-N.J., of the Foreign Relations Committee speak to ABC News Chief Global Affairs correspondent Martha Raddatz, Sunday on "This Week." And the powerhouse roundtable debates all the week's politics, with ABC News' George Will; Rep. David Schweikert, R-Ariz.; Democratic strategist and ABC News contributor Donna Brazile; NPR "Morning Edition" host Steve Inskeep; and New Republic owner and publisher Chris Hughes, who interviewed President Obama for an Oval Office exclusive on newsstands next week. Plus, "Zero Dark Thirty" screenwriter Mark Boal and "Blackhawk Down" author Mark Bowden discuss the controversy over the Oscar-nominated film. Tune in Sunday: http://abcnews.go.com/thisweek

THE ROUNDTABLE:

ABC's MICHAEL FALCONE: Although Louisiana Gov. Bobby Jindal is already the subject of speculation as a potential GOP presidential contender four years from now, if you ask him, it's way too soon for speculating. "Any Republican that's thinking about running for president in 2016 needs to get his head examined," Jindal told reporters after delivering a dinner speech at the Republican National Committee winter meeting last night. He added, "We've got a lot of work between now and the next midterm elections. ? We've got to get the Republican Party back on track." In his remarks to the gathering, he also offered some tough medicine for the GOP, including this piece of advice: "We must stop being the stupid party. It's time for a new Republican party that talks like adults. It's time for us to articulate our plans and visions for America in real terms. We had a number of Republicans damage the brand this year with offensive and bizarre comments. We've had enough of that." Whether or not Jindal ultimately emerges as a top presidential contender, look for him to be a major presence, not just in Louisiana, but around the country as well as a key figure in helping the party chart its course forward.

ABC's Z. BYRON WOLF: There's a bit of irony - if that's the right word - in the fact that it took fewer than 12 hours for senators to hatch, agree, and vote on a deal to tweak filibuster rules. The filibuster technically allows the minority to engage in unlimited debate. But changing the filibuster got barely any debate at all. Then again, the filibuster didn't really change. The deal passed overwhelmingly last night creates new rules to grease the arcane and usually slow-moving cogs of the "World's Greatest Deliberative Body." But 40 votes can still block anything. Would-be reformers say these rule changes are weak sauce. But it may represent a recognition in the Senate that things aren't really working the way they're supposed to.

ABC's RICK KLEIN: Can we get a round of applause for being quiet? Mitt Romney is in Washington today, of course a few days later than he wanted to have been in town, and on a far more temporary basis. What's remarkable, though, is how silent he's been in the 10-plus weeks since he didn't win the presidency. Save for a phone call with supporters that he didn't expect to come out - the one about the "gifts" that supposedly helped President Obama win a second term - we haven't heard Romney's voice at all since the election. He was not invited to the inauguration, and without a federal office he's holding, there would have been no reason for him to attend. There will be a time when Romney has something meaningful to contribute to the public dialog on, or something to reflect on or unburden himself with. In the meantime, until he feels like he has something to add, silence is worth savoring.

BUZZ

with ABC's Chris Good ( @c_good)

JOE BIDEN'S 'FIRESIDE HANGOUT': GUNS, EARTHQUAKES. ABC's Arlette Saenz reports: Sitting in front of a fireplace, Vice President Joe Biden participated in his first ever Google+ "Fireside Hangout" Thursday in his most recent attempt to push the administration's gun policy agenda, one that he said is focused on "gun safety" not "gun control." "I don't view it as gun control. I view it as gun safety," Biden said in a Google+ "Fireside Hangout" on gun violence. ? "My view is that it is totally a guarantee, not negotiable, that I'm able to own a weapon for sporting purposes as well as for my own protection, but there should be rational limits on the type of weapon" needed for protection or sports, he said. ? the vice president doled out a bit of advice for how to protect oneself after a deadly earthquake, telling one questioner a shotgun is more effective than an assault weapon. "If you want to keep people away during an earthquake, buy some shotgun shells," Biden said. http://abcn.ws/WpFAfn

PANETTA MAKES IT OFFICIAL: WOMEN WILL BE ABLE TO SERVE IN COMBAT. From ABC's Chris Good, Luis Martinez, and Mary Bruce: Women will soon be able to serve in combat, as things officially changed with the stroke of a pen today at the Pentagon. At a joint news conference, Secretary of Defense Leon Panetta and Joint Chiefs of Staff Charman Gen. Martin Dempsey signed a memorandum rolling back a 1994 directive prohibiting women from doing so. "They serve, they're wounded, and they die right next to each other," Panetta said of women and men in the military. "The time has come to recognize that reality. "If they're willing to put their lives on the line, then we need to recognize that they deserve a chance," Panetta said ? The change won't be immediate, however. While Panetta announced that thousands of new positions will now be open to women, he has asked the military branches to submit plans by May on how to integrate women into combat operations. He set a January 2016 deadline for branches to implement the changes, giving military services time to seek waivers for certain jobs. http://abcn.ws/WRuVuZ

OBAMA APPLAUDS THE MOVE. Obama hailed the move in a written statement. "Today, by moving to open more military positions - including ground combat units - to women, our armed forces have taken another historic step toward harnessing the talents and skills of all our citizens," he said. "This milestone reflects the courageous and patriotic service of women through more than two centuries of American history and the indispensable role of women in today's military," Obama said. "As commander in chief, I am absolutely confident that-as with the repeal of 'Don't Ask, Don't Tell'-the professionalism of our armed forces will ensure a smooth transition and keep our military the very best in the world," the president said. "Today, every American can be proud that our military will grow even stronger with our mothers, wives, sisters and daughters playing a greater role in protecting this country we love." http://abcn.ws/WRuVuZ (h/t ABC's Mary Bruce)

REPUBLICAN PARTY SET TO RE-ELECT CHAIR: Members of the Republican National Committee who have gathered for their winter meeting in Charlotte, N.C. today are poised to re-elect current Chairman Reince Priebus for another two-year term this afternoon. Priebus, the 40-year-old Wisconsin native who was elected to head the party in 2011, did not preside over a period that saw a winning Republican presidential candidate, but he did lead his party out of a deep hole of debt - more than $20 million left by former RNC Chairman Michael Steele. Chairman Priebus plans to deliver a speech to the meeting today and one of his big themes will be about how Republicans need to stay on a campaign footing at all times."Simple outreach' a few months before an election will not suffice," Priebus plans to tell the gathering, according to excerpts of his remarks. "In fact, let's stop talking about 'reaching out' - and start working on welcoming in. Political support is cultivated over time - not collected on Election Day." Chairman Priebus will also issue this call: "To those who have left the party, we want to earn your trust again. To those who have yet to join us, we welcome you-with open doors and open arms."

POST-ELECTION DATA WAR: TECHIES VS. POLS. The Verge's Ben Popper reported this week that Democratic operatives and Obama's tech team are fighting over what to do with the code that helped elect Obama: "[I]n the aftermath of the election, a stark divide has emerged between political operatives and the techies who worked side-by-side. At issue is the code created during the Obama for America (OFA) 2012 campaign: the digital architecture behind the campaign's website, its system for collecting donations, its email operation, and its mobile app. When the campaign ended, these programmers wanted to put their work back into the coding community for other developers to study and improve upon. Politicians in the Democratic party felt otherwise, arguing that sharing the tech would give away a key advantage to the Republicans. Three months after the election, the data and software is still tightly controlled by the president and his campaign staff, with the fate of the code still largely undecided. It's a choice the OFA developers warn could not only squander the digital advantage the Democrats now hold, but also severely impact their ability to recruit top tech talent in the future." http://bit.ly/UPNLDg

WHITE HOUSE, CONGRESSIONAL HISPANIC CAUCUS TO MEET ON IMMIGRATION. The Huffington Post's Elise Foley reports: "The White House will meet on Friday with members of the Congressional Hispanic Caucus to discuss immigration reform. The meeting, one of many between President Barack Obama's administration and Hispanic members of Congress, adds steam to Obama's inaugural pledge to push for immigration reform. Members of the Congressional Hispanic Caucus have provided guidance for the president in the past. The president's schedule for Friday says he will be at the White House in meetings, but does not specify whether he will speak with Hispanic Caucus members. The meeting was confirmed to The Huffington Post by Democratic sources not authorized to speak on the record about the meeting." http://huff.to/10W3mHx

WHY IS HILLARY CLINTON WEARING GLASSES? If you noticed the glasses Hillary Clinton wore during her appearances on the Hill to talk about Benghazi, ABC's Dana Hughes reports: "She'll be wearing these glasses instead of her contacts for a period of time because of lingering issues stemming from her concussion. With them on she sees just fine. In fact, she got a kick out of the above when she saw them crystal clear," Clinton spokesman Philippe Reines has confirmed.

TOM TANCREDO SAYS HE'LL SMOKE POT. Former congressman Tom Tancredo says he'll honor a bet to smoke marijuana. In a video to promote Colorado's Amendment 64, which legalized the personal use of marijuana in the state, Tancredo reportedly pledged to smoke pot if the measure passed. Tancredo, a self-professed non-marijuana-smoker, supported the measure. "Look, I made a bet with the producer of the film that if Amendment 64 passed (I did not think it would) that I would smoke pot," Fox News quotes Tancredo as saying. "I will therefore smoke pot under circumstances we both agree are legal under Colorado law. Hey, it's better than having to do a stupid dance as [Denver] Mayor [Michael] Hancock must perform as a result of losing a bet on the Broncos beating the Ravens." http://abcn.ws/14aH2su

MEMO TO FLIES: DON'T BUG OBAMA! Something was bugging President Obama yesterday as he nominated Mary Jo White to head the Securities and Exchange Commission and re-nominated Richard Cordray to lead the Consumer Financial Protection Bureau, ABC's Mary Bruce notes. "This guy is bothering me here," the president said, as he repeatedly swatted at a large black fly buzzing near his face. Given Obama's reputation, it was a bold move for the insect to spar with the "swatter in chief."During a June 2009 CNBC interview, Obama famously killed a fly with a single smack. "Get out of here," the president ordered of the fly before the interview began. When the directive was ignored, Obama waited for the fly to land on his hand and then, "slap," swiftly ended its life with a single blow. "That was pretty impressive, wasn't it?" Obama proudly declared. "I got the sucker." (PETA later condemned the move). The fly in the State Dining Room today was lucky enough to avoid a similar fate. http://abcn.ws/11UGiYQ
